Izvleček:Številne študije potrjujejo antidepresivno, antivirulentno, protibakterijsko in protitumorsko delovanje ekstraktov šentjanževke (Hypericum perforatum L.). Razvijali smo optimalni sistem za učinkovito ekstrakcijo po Soxhletu, ki bo služila za izolacijo hipericina kot glavne bioaktivne komponente iz posušenih cvetov in listov šentjanževke. Z metodo difuzijskih antibiogramov na vidni svetlobi smo raziskovali protimikrobni učinek etanolnih ekstraktov na po Gramu pozitivne (Listeria monocytogenes, Staphylococcus aureus, Staphylococcus epidermidis) in po Gramu negativne bakterije (Pseudomonas aeruginosa, Acinetobacter baumannii). Kot najbolj optimalna se je pokazala ekstrakcija iz posušenih cvetov v etanolu, med 15 in 20 ekstrakcijskimi cikli. Vsebnost hipericina v etanolnih ekstraktih cvetov je bila približno 4-krat večja kot v listih. Rezultati biološke aktivnosti v teh pogojih v nasprotju z nekaterimi študijami niso pokazali protimikrobne aktivnosti.
There is a growing demand for the medical plant Hypericum perforatum L. as it has been shown to possess various biological activities such as antidepressant, antiviral, antitumor, antioxidant and antibacterial effects. The aim of the experimental work was to develop the most optimum and efficient extraction procedure for the production of hypericin, a main bioactive compound, from dried flowers and leaves. With disk diffusion assay the antibacterial activity of ethanolic extracts on gram positive (Listeria monocytogenes, Staphylococcus aureus, Staphylococcus epidermidis) and gram negative bacteria (Pseudomonas aeruginosa, Acinetobacter baumannii) under the presence and absence of visible light was investigated. The results indicate that the best solvent for the effective extraction of hypericin is ethanol and that the hypericin content in extracts is decreasing with decreased solvent polarity. The content of hypericin per dry plant mass is significantly higher in flowers in comparison with leaves (4 times), but it was not even detected in the roots. The most optimum number of extraction cycles was found to be somewhere between 15 and 20 cycles. Contrary to some studies, results of disk diffusion assays did not show any signs of antibacterial activity in any of the tested samples of gram positive nor gram negative bacteria.
